what is also possible is

TRANQVILLITAS VIRTVS SAPIENTIAQVE

or the way Matthaeus did it - as an asyndeton (very common actually)

TRANQVILLITAS VIRTVS SAPIENTIA

virtus for fortitudo is fine of course

Why is a V used instead of a U?
are they the same or completely different?
i'm thinking of getting a tattoo that says Serenity, Courage and Wisdom in Latin
that is why i'm here getting help as I have no basic knowledge of the language.
So which is the best way for me to phrase this sentence in Latin?

(thanks for your help people, i really appreciate it)

The Romans had only the letter V. These days we usually distinguish u and v when writing Latin in lowercase, but the Romans didn't have lowercase letters either.

Lowercase letters (and by extension the distinction between "u" and "v" and later "w") are a Medieval invention.

ok may i ask one last question :)
tranquillitas is Serenity right?
so what is Amini in "Tranquillitas Amini" stand for?

and, If i just want to state these three words "Serenity, Courage, Wisdom" in this format "Serenity Courage Wisdom"
without any 'and' or ','
what would be the right way to write this in Latin?
"TRANQVILLITAS VIRTVS SAPIENTIA" <---- is correct right?

no, the double-l is correct. tranquillus is some sort of diminutive formed from trans + quies, hence the geminate l

Ooh dang, you're right. I should have stopped at animi.

Tranquillitas can indeed suggest tranquility of mind even without animi. I think that's what shawamoskoh is asking. With animi it's of course more explicit.
